Rechrom-Plasminogen

Reagent kit for the photometric determination of plasminogen (Rechrom-Plasminogen) according to TU 9398-270-05595541-2010. Designed for the quantitative determination of plasminogen activity in blood plasma using a chromogenic method, both manually and on automatic analyzers.

Plasmin, an active enzyme involved in both primary and secondary fibrinolysis, circulates in the plasma in large quantities as its inactive precursor, plasminogen. Plasminogen can be activated to plasmin by various pathways, with the most important being activation by natural tissue activator synthesized and released by endothelial cells. Plasminogen can also be activated by external activators such as streptokinase from hemolytic streptococcus and urokinase, a natural activator from the kidneys. Plasminogen consumption is observed in both primary and secondary fibrinolysis. Secondary fibrinolysis, associated with disseminated intravascular coagulation, is the most important cause of plasminogen consumption. On the other hand, primary fibrinolysis, involving only the fibrinolytic mechanism, also leads to rapid consumption of the circulating enzyme.

Principle of the method: The method for determining plasminogen activity in a plasma sample is based on its ability to form a complex with streptokinase, which hydrolyzes a chromogenic peptide substrate. The amount of released para-nitroaniline (pNA) is directly proportional to the plasminogen activity in the plasma sample. The process proceeds according to the following scheme: Plasminogen + Streptokinase (excess) ⇒ Complex Complex + Peptide-pNA ⇒ Peptide + pNA (yellow) A calibration curve is constructed using plasma calibrator with a known plasminogen activity (A). Optical density values obtained for each dilution of the plasma calibrator are plotted on the Y-axis, and plasminogen activity in % is plotted on the X-axis on a linear scale. Using the calibration curve and the optical density value of the test sample, the plasminogen activity is determined.
